The instructions with my Amazon cheapies say to dip the OPKs in room-temperature urine. Does anyone actually wait for that and does it make any difference?

Never heard that. I wouldn't wait. The only thing that people suggested to me on here that I did and found more successful was to use smu for opks and pregnancy tests too actually! It made my cheapies much more successful/accurate I feel.

Never did that either. I used my OPKs mainly between 530-6pm when I got home from work.

Never waited for my pre to cool off, I typically did them after a few hr hold in the evening.
