Has anyone here used the Up & UP OPK strips from Target? Just the little sticks that you dip. The instructions say to use FMU and I always heard to test later in the day. Any insight?

I didn't use that particular brand, but I don't think brand really matters. I've read that the LH is typically first released in the body in the morning, so afternoon will give you the earliest positive opk. However, most surges can be detected for 24-ish hours, so you should be able to get a positive first thing the next morning too.
I typically took mine in the afternoon/evening.

I think I'll start testing in the afternoon and morning to see if anything changes.
